uhrf1^hi3020 is a null allele. (A) Schematic of the uhrf1 genomic locus with the approximate position of the proviral insert and viral or exon-specific primers used for RT-PCR analyses indicated. (B) RT-PCR using the indicated primer sets reveals an absence of uhrf1 transcripts in recessive mutants. Beta actin is used as a loading control. (C) Western blot using protein extracts derived from whole, de-yolked 5 dpf wild-type sibling and uhrf1 mutant embryos. Anti-Uhrf1 recognizes a band of approximately 85 kDa in wild-type samples, which is absent in those from uhrf1 mutants. anti-Hdac1 is used as a loading control.
