Expression of camk2b2. (a) The amino acid sequence of β2 CaMK-II, predicted from its gene sequence (camk2b2) is shown containing only the non-alternative (II and VII) variable region domains. Underlined residues are unique to and therefore diagnostic for β2 CaMK-II. (b) β2 CaMK-II splice variants were RT-PCR amplified from 0 to 84 hpf whole embryos, cloned and sequenced. Four variants were identified based on alternative domain usage; none utilizes the putative nuclear targeting domain III. (c) camk2b2 expression was assessed by in situ hybridization at the 8 and 512-cell, 8- and 16-somite and at indicated hours (24, 34, 36, 48 and 72) post fertilization (hpf). Anterior is to the left except for the head on view of the heart at 36 hpf. Arrow heads indicate fin buds, asterisks indicate heart, v = ventricle, a = atrium, scale bar = 1 mm. |
