Argonaute family tree and ziwi expression
(A) Evolutionary tree drawn derived from the alignments of sequences of different Argonaute family members. The Argonaute family is divided to two distinct subclasses. The somatic AGO branch is shown in black and the germ line specific PIWI branch is depicted in red, which all share a PAZ and a PIWI domain. Zebrafish PIWI proteins are Ziwi (ENSDARG00000041699) and Zili (Piwil2) (ENSDARG00000035614).
(B) Fluorescent in situ hybridization showing that ziwi is expressed in germ cells in 40 day-old females (top row) and adult males (bottom row). Green: Vasa protein; Red: Ziwi mRNA. The right most panels show a non-fluorescent in situ hybridization on adult ovary and testis.
(C) Expression of ziwi in wild-type testis, testis from Dead-end morpholino injected fish, which do not contain germ cells, and testis from ziwi-/- mutant fish. (a) RT PCR for the indicated genes. (b) -RT control. Elf1 is a house keeping gene.
