Generation of zebrafish transgenic lines expressing wild type and mutant human TDP-43. (A) The transgene constructs consist of the human wild type (TDP-43WT) or the G348C mutant TDP-43 (TDP-43G348C) cDNA fused with 6 myc-tag under the control of the hsp70-like promoter (hsp70l). The constructs also contained eGFP driven by the cardiac myosin light chain (cmlc2) promoter. (B) Representative picture of the embryos used for all experiments. (C) Western blot showing expression of the human transgene in 2 days post-fertilization (dpf) embryos following a 1 h heat shock at 38.5°C using an anti-myc antibody. |
