RUNX1-Evi-1 reprogrammed lineage-specific hematopoietic transcription factors. Scl (a), lmo2 (b) and gata2 (c) were detected by qRT-PCR in WT, and Tg(RE:HSE:EGFP) F2 generation embryos at 12 hpf, 18 hpf and 24 hpf. Gata1 (d), pu.1 (e), mpo (f), runx1(g), and c-myb (h) expressed in WT, Tg F2 zebrafish at 1 dpf, 3 dpf, 7 dpf and 60 dpf. Compared with WT, scl, gata2, pu.1, and c-myb were up-regulated, while gata1, mpo, and runx1 were down-regulated in Tg fish. In situ hybridization of scl (i-j), lmo2 (k-l), gata2 (m-n), and gata1 (o-p) at 24hpf and mpo (q-r) at 30hpf in WT and Tg F2 embryos demonstrated the same tendency. *P < 0.05; **P < 0.01 |
