Expression folr1 mRNA from the Cleavage to Gastrula stages of zebrafish development. We observed the presence of the folr1 transcript throughout these early stages of development suggesting that folr1 is also maternally deposited and then upregulated at the maternal to zygotic transition. The expression of folr1 becomes restricted during and after segmentation. We observed folr1 expression become restricted to regions of known cell proliferation during and after somitogenesis. Of interest is the expression in the pectoral fin at 2dpf & the cloaca at 4dpf. Folate targeted compounds are selectively taken up and maintained in the embryo. Brightfield and fluorescence representative images of wild-type fish exposed to either BSA conjugated to rhodamine dye or folate conjugated to BSA and rhodamine dye. After 24 h exposure to BSA based compounds, embryos were washed and transferred to standard embryo media. Exposed embryos were then imaged at 24 and 48 h after being transferred to standard embryo media and shown above.
